New Drug Modalities Project

March 25, 2024

JASCO is running the New Drug Modalities Project, which focuses on creating dedicated products and applications for research and development in antibody and nucleic acid-based drug market.

This topic introduces cutting edge solutions and applications, including circular dichroism spectrometers and infrared spectrometers, capable of providing insights into the secondary and tertiary structures of antibodies and nucleic acids, as well as Raman spectrometers for evaluating higher-order structures. Additionally, the utilities of spectrofluorometers, UV-visible spectrophotometers and size-exclusion chromatography (SEC) are also introduced.

Topics

Statistical Evaluation of Structural Changes in Biopharmaceuticals - qHOS Program -

Since circular dichroism spectroscopy can obtain information regarding the higher-order structure of proteins in quick, this technique is used for R&D and quality control of biopharmaceuticals. However, the significant differences in CD spectra is judged by empirical and visual assessments.

The qHOS program offers a solution by employing statistical tests to identify significant differences among multiple spectra. This enables objective evaluation of the equivalence of higher-order structures in biopharmaceuticals.

Highly Accurate Secondary Structure Estimation of Protein - BeStSel CFR Program

Recently, Micsonai et al. developed the BeStSel algorithm that can accurately estimate the secondary structure composition from the CD spectrum by taking into account the parallel-antiparallel orientation of the β-strands and the twist of the antiparallel β-sheets. Spectra Manager™ 2.5 BeStSel CFR offers a control and analysis platform for CD spectrometers, which is compatible with GxP and satisfying computer system validation (CSV), electronic record/electronic signature (ER/ES), and data integrity (DI) for practice ALCOA+.

Automation of circular dichroism spectra measurement - HTCD Plus

Since circular dichroism spectrometer can acquire higher-order structures of protein and nucleic acid under various solvent conditions easily and rapidly, this is used for various situations such as R&D and quality control. The HTCD Plus (High-throughput Circular Dichroism Measurement System) enables to acquire the data of multiple samples effectively by automating the measurement and cleaning process.

Tm Analysis of Nucleic Acids

V-700 series UV-Visible spectrophotometer has capability of Tm measurement by combining with Peltier cell holder and the dedicated program, which includes automatic annealing function.

The air-cooled Peltier cell holder allows temperature control ranging from 0 to 100 °C without water circulator, simplifying temperature-dependent measurements.

For multi-sample measurements, the water-cooled Peltier cell holder PAC-743/PAC-743R can measure up to 8 samples with ease.
